P>The objective of this study was to complement published hake (Merlucius merlucius) sperm motility characteristics by obtention of data more specifically dependent on flagella: beat frequency, flagellar wave amplitude, wave length, number of waves along the length of the flagellum and curvature of the wave. The changes of these parameters are described in relation to the time period elapsed since activation are described using high resolution video images of flagella while quantifying the progression during the swimming period from the activity initiation to full cessation of movement several minutes later. Effect of age and environmental conditions on the movement characteristics of Pacific oyster (Crassostrea gigas) trochophores ArchiMer

The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. Improvement of chilled seabass sperm conservation using a cell culture medium ArchiMer

The sperm of seabass is very fragile and it quickly loses its ability to fertilize after collection either if kept undiluted or in classic saline media. In order to avoid cryopreservation when only short conservation is required, the process of sperm management including sperm collection, sperm dilution rate in storage medium and storage medium composition, was subject to experimental trials. A concentration of 20% urine generated a low pH of seminal fluid, and it immediately altered the motility ability. However, pH did not seem to be the key agent of motility prevention since sperm dilution in Leibovitz culture medium (L15) or in classic saline medium both presenting a similar low pH (7.3) did not affect motility. Marathon vs sprint racers: an adaptation of sperm characteristics to the reproductive strategy of Pacific oyster, turbot and seabass ArchiMer

This work assesses the present knowledge on Pacific oyster sperm biology in comparison to two marine fish species (turbot and seabass) whose sperm characteristics are well described. Sperm morphology mainly differs by the presence of an acrosome in Pacific oyster which is absent in both fish species. In turbot as in Pacific oyster, a sperm maturation process along the genital tract is observed. Sperm motility is triggered by changes in osmolality for seabass and turbot and in pH for Pacific oyster. However, complementary factors are involved to maintain sperm immotile in the genital tract. P>The objective of this paper is to characterize some of the sperm motility parameters in European hake (Merluccius merluccius), which is considered to be a species with aquaculture potential. The total ATP, ADP and AMP concentrations were determined using high-performance liquid chromatography on hake sperm samples collected during the winter-early spring in the Bay of Biscay (France) (n = 22) and on hake sperm samples collected during the summer-early autumn in waters off Western Norway (n = 5). Sperm features of captive Atlantic bluefin tuna (Thunnus thynnus) ArchiMer

P>The present study aimed to establish some basic characteristics of Atlantic bluefin tuna sperm from captive mature males, treated or untreated by gonadotropin releasing hormones agonist (GnRHa). Intratesticular milt was collected from treated and untreated fish (mean weight +/- SD: 122.9 +/- 29.2 kg, n = 21). There was no significant effect of GnRHa treatment on GSI (1.33 +/- 0.70%, n = 21) or on sperm concentration (3.8 +/- 1.3 x 1010 spermatozoa ml-1, n = 21) estimated by optical density at 260 nm.
